Obtain a highly purified, homogeneous sample at a concentration and thickness appropriate for TEM or cryoEM.
Preserve the structure of the specimen for examination in a TEM at the desired resolution.
For cryoEM this involves rapid freezing or vitrification, which preserves the specimen in a native, hydrated state.
Mount the specimen on grids with a thin carbon film or, ideally for cryoEM, suspended in the holes of a
holey support film to reduce background noise.
